C'est un inhibiteur spécifique de Cdk4 et Cdk6 et se lie au complexe Cdk-cycline phosphorylé. Une perturbation de cette voie est couramment observée dans le cancer. Le p16 est perdu dans la majorité des lignées cellulaires tumorales et dans la plupart des tumeurs primaires. Il n'est pas exprimé dans le mélanome. Les lames ont été incubées avec une solution de DAB jusqu'à ce qu'une coloration brune soit visible et avec de l'hématoxyline pendant quelques minutes.
