Le papier ROTITEST® Phosphatase, imprégné de réactif de détection, est découpé à une extrémité et laissé à imbiber en le trempant dans un échantillon du lait à analyser. Pendant l'incubation à 37 °C pendant 2 heures au maximum, une coloration jaune indique la présence de phosphatase alcaline, et donc une pasteurisation inefficace du lait.
La sensibilité du système de test de la phosphatase est de 0,5 % de lait cru dans le lait pasteurisé. Ce système de test ne fournit pas de données quantitatives.
