Dit antilichaamportfolio bevat een breed scala aan weefsel- en tumorspecifieke primaire antilichamen voor fundamenteel onderzoek, productontwikkeling en de analyse van diverse biologische vraagstukken. Perilipinen vormen een familie van fosfoproteïnen. De dominante vormen in adipocyten, PLIN1 A en B, ontstaan door alternatieve RNA-splicing uit een enkel gen, waarbij polypeptiden van respectievelijk 57 kDa en 46 kDa worden gegenereerd. De N-terminus blijft echter ongewijzigd. Het antilichaam reageert specifiek met alle PLIN1-varianten die zich bevinden aan het oppervlak van intracellulaire opslag-lipidedruppels, aanwezig in bijv. de bijnier, talgklier, adipocyten van wit en bruin vetweefsel en gekweekte cellen zoals 3T3-L1-adipocyten en gekweekte steroïdogene bijnierschors- en Leydig-cellen. Het is tevens een nuttige pathologische marker voor steatogenese, bijv. in de lever. Het vertoont geen kruisreactiviteit met adipofiline (ADRP, ook wel PLIN2 genoemd) of TIP47 (ook wel PLIN3 genoemd) eiwitten (of andere leden van de PLIN/PAT-familie, bijv. MLDP of OXPAT/PAT-1, ook wel PLIN5 of LSDP5 genoemd). Geteste reactiviteit op gekweekte cellijnen: diverse humane carcinoom-cellijnen; 3T3-L1-adipocyten.

Immunocytochemie (ICC): assay-afhankelijkImmunohistochemie (IHC) - bevroren: 1:200 (0,5 μg/ml)Immunohistochemie (IHC) - paraffine: 1:200 (0,5 μg/ml, magnetronbehandeling aanbevolen)

IHC-analyse van humane adipocyten in wit vetweefsel van de tumor-micro-omgeving met gebruik van het anti-Perilipine 1 (N-terminus) antilichaam. IHC werd uitgevoerd op in formaline gefixeerde, in paraffine ingebedde coupes. De monsters werden gedeparaffineerd met xylol en ethanol, gevolgd door hitte-geïnduceerde antigeen-retrieval met 10 mM citraatbuffer. Na voorbereiding werd het weefsel 20 min geblokkeerd met normaal serum bij RT. Het primaire antilichaam anti-Perilipine 1 (N-terminus), PERI 112.17 (cat.nr. 690156) werd verdund in PBS (antilichaamconcentratie 500 ng/ml) en 's nachts geïncubeerd bij 4 °C. Het secundaire antilichaam ImmPRESS HRP anti-muis IgG werd 20 min geïncubeerd bij RT. De objectglaasjes werden met DAB-oplossing geïncubeerd totdat een bruine kleuring zichtbaar was en vervolgens enkele minuten met Haemalaun. De 10x-opname werd verkregen door middel van microscopie (met dank aan J. Hess, Universitair Ziekenhuis Heidelberg).
