Het assortiment ROTI®Antibiotic Discs is ontwikkeld voor de kwalitatieve analyse van de gevoeligheid van bacteriën of schimmels voor antimicrobiële reagentia door middel van in-vitro-diffusiemethoden.
Een verse starterscultuur wordt ingesteld op een celdichtheid van 1×106-5×106 kve/ml (volgens de McFarland-standaard 0,5) en gelijkmatig aangebracht op een kweekbodemplaat met Mueller-Hinton-agar (Art.nr. X926; voor schimmelculturen 2 % glucose + 0,5 μm methyleenblauw toevoegen). De antibioticaschijfjes worden met een steriele pincet of een dispenser (Art.nr. 1505) individueel op de plaat aangebracht.
Na een geschikte incubatieperiode wordt de grootte van de remmingszone rond de schijfjes gemeten. De diameter van de remmingszone wordt beoordeeld met behulp van gestandaardiseerde waarden in de beschikbare normen en databases en is een indicator voor de gevoeligheid van de cultuur voor het antibioticum.
Het wordt aanbevolen om 2 % glucose en 0,5 µg methyleenblauw aan Mueller-Hinton-agar (Art.nr. X926) toe te voegen.
Elk ROTI®Antibiotic Discs Amphotericin B (AP)-schijfje bevat amfotericine B in een concentratie van 30 μg en is aan beide zijden bedrukt met "AP 30". De diameter van de ROTI®Antibiotic Discs is 6 mm.
