Laadbuffer met de fluorescerende kleurstof SYBR® Green I voor het kleuren van dubbelstrengs nucleïnezuur in agarose- en polyacrylamidegels.

ROTI®Load DNAstain SYBR® Green is eenvoudig in gebruik en verkort de tijd die nodig is voor gelanalyse aanzienlijk. De aanwezige fluorescerende kleurstof SYBR® Green I detecteert tot 0,01 ng per band en is daarom 20 maal gevoeliger dan ethidiumbromide. De integriteit van het DNA wordt door SYBR® Green I niet beïnvloed; daarom kan uit de gel geëlueerd DNA direct worden gebruikt voor standaard downstream-toepassingen, zoals bijv. ligatie, PCR, transformatie, transfectie enz., zonder storende effecten. SYBR® Green I bindt aan de kleine groeve van het DNA en intercaleert in het DNA. Het kan worden geëxciteerd door UV- en blauw licht, waardoor het volledig compatibel is met standaard UV-transilluminatoren en met moderne blauw-licht-verlichting (zie ook ROTIPHORESE®PROfessional runVIEW, art.nr. 4849.1). Bij excitatie zendt het aan nucleïnezuur gebonden SYBR® Green I een felgekleurde groene fluorescentie (521 nm) uit die kan worden gedocumenteerd met alle gebruikelijke breedband ethidiumbromide- of SYBR® Green-fotofilters. De afbeelding toont 50 ng DNA per band, of respectievelijk 75 ng (1000 bp en 2,5 kb). Elke baan is geladen en gelijktijdig gekleurd met 1 µl ROTI®Load DNAstain SYBR® Green.
