Trimethoprim inhibits the enzyme dihydrofolate reductase in bacteria, which blocks the synthesis of tetrahydrofolic acid. As tetrahydrofolic acid is essential for the production of nucleic acids and proteins, this inhibition leads to the bacteria no longer multiplying and eventually dying off.
