p16 (CDKN2A, p16Ink4A) est un régulateur clé du cycle cellulaire et est impliqué dans le contrôle du cycle cellulaire et la sénescence cellulaire. C'est un inhibiteur spécifique de Cdk4 et Cdk6 et se lie au complexe Cdk-cycline phosphorylé. Une perturbation de cette voie est couramment observée dans le cancer. Le p16 est perdu dans la majorité des lignées cellulaires tumorales et dans la plupart des tumeurs primaires. Il n'est pas exprimé dans le mélanome. Dans les carcinomes dus à une infection par le VPH (virus du papillome humain), le p16 est souvent surexprimé. L'anticorps est particulièrement utile pour l'immunoprécipitation. L'épitope a été localisé dans les 15 résidus d'acides aminés de l'extrémité C-terminale de la protéine p16.

Analyse IHC d'un carcinome épidermoïde humain à l'aide d'un anticorps anti-p16. L'IHC a été réalisée sur des coupes fixées au formol et incluses dans la paraffine. Les échantillons ont été déparaffinés avec du xylol et de l'éthanol, suivis d'une récupération d'antigène induite par la chaleur avec un tampon citrate 10 mM. Après préparation, le tissu a été bloqué avec du sérum normal pendant 20 min à température ambiante. L'anticorps primaire anti-p16 souris monoclonal, DCS-50 (N° de cat. 690074) a été dilué dans du PBS (concentration d'anticorps 2,5 µg/ml) et incubé à 4°C pendant la nuit. L'anticorps secondaire ImmPRESS HRP anti-IgG de souris a été incubé pendant 20 min à température ambiante. Les lames ont été incubées avec une solution de DAB jusqu'à ce qu'une coloration brune soit visible et avec de l'hématoxyline pendant quelques minutes. L'image 20x a été acquise à l'aide d'un microscope (avec l'aimable autorisation de J.Hess, Hôpital universitaire de Heidelberg).
Analyse Western blot du lysat de HeLa avec l'anticorps anti-p16. L'analyse Western blot a été réalisée sur 20 µg de lysat de HeLa. Les cellules ont été lysées avec un tampon RIPA. La membrane PVDF a été bloquée avec 5% de lait sec dans du PBST (PBS + 0,1% Tween 20) pendant 1 h à température ambiante. L'anticorps primaire anti-p16 souris monoclonal, DCS-50 (N° de cat. 690074) a été dilué dans le tampon de blocage (concentration d'anticorps 1 µg/ml) et incubé pendant 1 h à température ambiante. L'anticorps secondaire anti-IgG de souris conjugué HRP a également été dilué dans le tampon de blocage (concentration d'anticorps 0,2 µg/ml) et incubé pendant 1 h à température ambiante. Les bandes ont été visualisées par détection chimiluminescente à l'aide du substrat Pierce™ ECL Western Blotting.
