Dans le système de buvardage semi-sec, il est possible d'utiliser aussi bien un système de tampon continu (tampon identique à l'anode et à la cathode) qu'un système de tampon discontinu (tampon différent à l'anode et à la cathode). Les performances de transfert des systèmes discontinus sont cependant généralement supérieures, car les deux tampons sont conçus séparément selon les besoins des deux électrodes. Le système de tampon discontinu ROTI®Blot 1 se compose de deux tampons différents, le tampon d'anode A (pH 7,8 ±0,1) et le tampon de cathode K (pH 8,5 ±0,1). ROTI®Blot 1 présente une efficacité de transfert bien meilleure qu'un tampon tris-glycine et peut être utilisé pour des protéines et des peptides de toute longueur.
La pile de transfert (blot stack) est préparée en imbibant les papiers de buvardage supérieurs avec le tampon de cathode et les papiers de buvardage inférieurs avec le tampon d'anode. Avec des temps de transfert légèrement plus longs, les protéines >100 kDa peuvent également être complètement transférées avec ROTI®Blot 1. Un manuel d'instruction détaillé est joint au produit.
