Dulbecco’s Modified Eagle Medium is de meest gebruikte modificatie van het basismedium volgens Eagle. DMEM bevat een hogere concentratie aan aminozuren en vitaminen, en bovendien ijzer (ijzer(III)nitraat). De basissamenstelling van ROTI®Cell DMEM-media komt overeen met de oorspronkelijke formulering volgens Dulbecco. De formulering met een hoog glucosegehalte is zeer geschikt voor het kweken van een breed scala aan zoogdiercellijnen en primaire cellen.
Over het algemeen wordt FBS toegevoegd aan DMEM-basismedia.
Deze modificatie van DMEM bevat stabiel glutamine en pyruvaat.
Stabiel glutamine is aanwezig als het dipeptide L-alanyl-L-glutamine. Vanwege de versterkte structuur is deze vorm van glutamine, in tegenstelling tot normaal L-glutamine, niet temperatuurgevoelig en valt daarom niet spontaan uiteen in toxische ammoniak. Cellen zijn in staat om deze dipeptidebinding af te breken en naar behoefte L-glutamine vrij te maken. De lage ammoniakconcentratie verbetert hierdoor de levensvatbaarheid en groei van de cellen. Bovendien wordt de concentratie L-glutamine constant gehouden. Stabiel glutamine is al aan sommige celkweekmedia toegevoegd, maar kan ook als afzonderlijke additieve oplossing worden gekocht, de ROTI®Cell Glutamine Solution.
