Le Minimum Essential Medium est une modification courante du Basal Medium selon Eagle, développée en 1959 pour la culture de cellules HeLa. Le MEM contient une forte concentration d'acides aminés, qui correspondent étroitement à la composition protéique des cellules humaines. La concentration de vitamines et d'autres nutriments essentiels est augmentée, permettant des périodes de culture plus longues entre les repas. Le MEM est l'un des milieux de culture cellulaire les plus utilisés et peut être utilisé pour une variété de cultures cellulaires en suspension et de cellules adhérentes.
Généralement, le FBS est ajouté aux milieux de base MEM.
Cette modification du MEM contient de la glutamine stable et des sels d'Earle, mais pas de pyruvate.
La glutamine stable est présente sous forme du dipeptide L-alanyl-L-glutamine. En raison de sa structure renforcée, cette forme de glutamine n'est pas sensible à la température, contrairement à la L-glutamine normale, et ne se décompose donc pas spontanément en ammoniac toxique. Les cellules sont capables de rompre cette liaison peptidique et de libérer de la L-glutamine selon leurs besoins. Ainsi, la faible concentration d'ammoniac améliore la viabilité et la croissance cellulaires. De plus, la concentration de L-glutamine est maintenue constante. La glutamine stable a déjà été ajoutée à certains milieux de culture cellulaire, mais peut également être achetée sous forme de solution additive séparée, la ROTI®Cell Glutamine Solution.
