Le phénol et le chloroforme provoquent la dénaturation des protéines qui s'accumulent dans l'interphase. L'alcool isoamylique empêche la formation de mousse et inhibe très efficacement les ARNases. Pour une séparation de phases nette, ROTI®Aqua-Phénol nécessite un pH ≤ 4,0 dans la phase aqueuse supérieure. Si nécessaire, la phase supérieure peut être acidifiée en ajoutant 1 à 2 gouttes de HCl 1 N.
