Europese Farmacopee 2.6.7 Mycoplasma's vereist validatie van de gevoeligheid en robuustheid ten opzichte van de monstermatrix. Bovendien moet de methode een prestatie laten zien die gelijk is aan of beter is dan die van de klassieke kweekprocedure. Omdat de meeste test- en celkweeklaboratoria bang zijn om mycoplasma te kweken, is een dergelijke vergelijking niet eenvoudig te realiseren. Microsart® Validatiestandaarden zijn niet infectieus en daarom veilig in gebruik. Ze worden getitreerd tot 10 CFU/ml, de gevoeligheidslimiet voor NAT-gebaseerde methoden zoals PCR ter vervanging van de traditionele kweekmethode. De mycoplasmacellen die worden gebruikt voor de productie van de Microsart® Validatiestandaard zijn referentiestammen met een lage passage, gekweekt in kweekbouillon zoals beschreven in EP 2.6.7. De culturen worden geoogst in de vroege logaritmische fase van de groei om een atypisch hoog percentage dode mycoplasmadeeltjes te voorkomen, direct getitreerd in kweekbouillon en uitgeplaat op Hayflick- en Frey-medium voor kwantificering op basis van kolonievormende eenheden (CFU).
