Minimum Essential Medium is een veelgebruikte modificatie van het Basal Medium volgens Eagle, ontwikkeld in 1959 voor het kweken van HeLa-cellen. MEM bevat een hoge concentratie aminozuren die nauw aansluiten bij de eiwitsamenstelling van menselijke cellen. De concentratie van vitaminen en andere essentiële voedingsstoffen is verhoogd, wat langere kweekperioden tussen de voedingsbeurten mogelijk maakt. MEM is een van de meest gebruikte celkweekmedia en kan worden ingezet voor diverse suspensiecelkweken en adherente cellen.
Doorgaans wordt aan MEM-basismedia FBS toegevoegd.
Deze modificatie van MEM bevat stabiel glutamine en Earle's zouten, maar geen pyruvaat.
Stabiel glutamine is aanwezig als het dipeptide L-alanyl-L-glutamine. Vanwege de versterkte structuur is deze vorm van glutamine, in tegenstelling tot normaal L-glutamine, niet temperatuurgevoelig en ontleedt daarom niet spontaan tot giftig ammoniak. Cellen zijn in staat om deze dipeptidebinding af te breken en naar behoefte L-glutamine vrij te maken. Hierdoor verbetert de lage ammoniakconcentratie de levensvatbaarheid en groei van de cellen. Bovendien wordt de concentratie L-glutamine constant gehouden. Stabiel glutamine is al aan sommige celkweekmedia toegevoegd, maar kan ook als afzonderlijke additieve oplossing worden aangeschaft, de ROTI®Cell Glutamine Solution.
