ROTI®Garose-Biotin en Streptavidine beads bieden de perfecte oplossing voor de isolatie van gebiotinyleerde en streptavidine-gelabelde moleculen in benchtop kolomchromatografie of in batch-procedures.

Met een waarde van Ka~10-14/M is de affiniteit van avidine en streptavidine voor biotine de sterkste niet-covalente binding tussen moleculen die in de biochemie bekend is. Efficiënte en zeer specifieke isolatie van gebiotinyleerde of avidine/streptavidine-gelabelde moleculen via affiniteitschromatografie is gebaseerd op deze specifieke affiniteit.
De matrix is gemakkelijk te verpakken en resulteert in zeer gelijkmatig verpakte kolommen. Elutie wordt uitgevoerd via 8 M guanidine-HCl of vóór het aanbrengen op de gel tijdens verhitting in de gel-laadbuffer. Er vindt vrijwel geen aspecifieke binding van niet-gelabelde moleculen plaats, zelfs niet tijdens zuivering uit ruwe extracten.

De matrix van ROTI®Garose-Biotin Beads bestaat uit 4% cross-linked agarose-beads. Biotine is aan de beads geïmmobiliseerd door middel van een spacer-arm en een covalente carboxy/amide-verbinding, wat niet alleen de lekkage van biotine tijdens elutie minimaliseert, maar ook de bindingscapaciteit verhoogt door reductie van sterische effecten. Er vindt vrijwel geen aspecifieke binding van niet-gelabelde moleculen plaats, zelfs niet tijdens zuivering uit ruwe extracten.
