ROTI®Pol, de serie DNA-polymerasen is de optimale keuze voor alle PCR-cyclingprotocollen, zoals uitgevoerd bij bijvoorbeeld analyse van kloonefficiëntie, voor gen-fishing, bij routinematige screeningprocessen, educatieve assays en nog veel meer. In combinatie met onze speciaal ontworpen buffers leveren de ROTI®Pol DNA-polymerasen een specifieke en reproduceerbare PCR-amplificatie met een breed scala aan PCR-templates.
Geoptimaliseerde voorgemengde 2x PCR-oplossing met het recombinante TaqS DNA-polymerase van de thermofiele bacterie Thermus aquaticus, dNTP's, MgCl2 en alle andere componenten die nodig zijn voor PCR (zonder primers en template-DNA), plus componenten voor directe gelelektroforese. ROTI®Pol TaqS Red-Mix (2x) wordt aanbevolen voor gebruik in alle standaard PCR-toepassingen die direct worden gevolgd door gelelektroforese. PCR-analyse met de ROTI®Pol TaqS Red-Mix (2x) mastermix vermindert niet alleen contaminatierisico's, maar is ook tijdbesparend, zeer reproduceerbaar en zeer eenvoudig voor te bereiden. ROTI®Pol TaqS Red-Mix (2x) kan worden toegepast op alle standaard Taq-gebaseerde cyclingprotocollen en is daarom de optimale keuze voor high-throughput PCR, zoals uitgevoerd bij bijvoorbeeld analyse van kloonefficiëntie of koloniescreening, voor routinematige screeningprocessen en voor studentencursussen. Dankzij de geoptimaliseerde samenstelling van de mastermix levert het TaqS-polymerase een specifieke PCR-amplificatie met een goede opbrengst bij een breed scala aan PCR-templates. ROTI®Pol TaqS Red-Mix (2x) is in staat om PCR-producten tot 3 kb te amplificeren met genomisch DNA en is geschikt voor gebruik met pure DNA-oplossingen, cDNA en bacteriële kolonies als templates. In 1% agarosegel migreert de inbegrepen rode kleurstof ongeveer even snel als een 1 kb DNA-fragment. Tijdens denaturatie bij Southern blotting kleurt de kleurstof geel bij een zure pH. Het TaqS-polymerase in de mastermix bezit zowel een 5’ → 3’ polymerase- als een 5’-flap-endonuclease-activiteit en genereert een 3’dA (adenine)-overhang die goed kan worden gebruikt voor TA-kloondoeleinden.
Respectievelijk 2 of 10 vials met elk 1 ml ROTI®Pol TaqS Red-Mix (2x) bevattend TaqS-polymerase, 0,4 mM van elk dNTP en 4 mM MgCl2 in 2x reactiebuffer met 0,02 % cresolrood.
