Populair substraat voor peroxidasen. DAB wordt gebruikt voor de biochemische en histologische detectie van endogene peroxidasen zoals catalase en cytochroom-oxidase. Bovendien is het het meest gebruikelijke substraat voor mierikswortelperoxidase (HRP) bij histochemische en cytologische assays. DAB wordt door HRP geoxideerd, waarbij een bruin neerslag wordt gevormd dat onoplosbaar is in waterige en organische oplosmiddelen. Dit neerslag kan in zichtbaar licht worden gedetecteerd en verbleekt niet bij langdurige opslag.
Werkconcentratie: 0,05-0,1 % (0,5-1 mg/ml) DAB in buffer (PBS, TBS, pH 7,0-7,6) met 0,01 % waterstofperoxide. Gebruik altijd een vers bereide werkoplossing. Voor oxidatie zijn hoge concentraties vers waterstofperoxide nodig.
