DEAE is het oudst bekende transfectiereagens. De methode van complexvorming van plasmide-DNA met diethylaminoethyldextraan werd in 1968 gepubliceerd door McCutchan. Positief geladen DEAE bindt negatief geladen DNA tot aggregaten, die vervolgens binden aan negatief geladen oppervlaktestructuren van celmembranen. Opname vindt plaats via endocytose. Deze DEAE-methode vereist slechts kleine hoeveelheden DNA, terwijl ze een hoge transfectie-efficiëntie van maximaal 30 % oplevert. De cytotoxische effecten veroorzaakt door DEAE staan echter alleen transiënte transfecties toe. De volgende cellijnen zijn goed transfecteerbaar: CV-1, BSC-1 en COS-cellen, en andere stabiele, ongevoelige cellijnen. Voorraadoplossing: 50 mg/ml in H2O. Autoclaveren.
