Trypsine is een eiwitsplitsend spijsverteringsenzym uit de pancreas dat behoort tot de serineproteasen en dat lysine en arginine aan de C-terminus splitst. Trypsine-oplossingen worden gebruikt voor het losmaken van cellen uit weefsel of celmonolagen. Ze zijn daarom essentieel voor routinetoepassingen bij het passageren van celculturen, voor celkulturexperimenten om een adequaat aantal cellen te verkrijgen of voor de dissociatie van primair weefsel. Als alternatief voor Trypsine kan de celvriendelijkere oplossing ROTI®Cell Accutase-oplossing worden gebruikt, die vrij is van componenten van zoogdieren.
De vereiste trypsineconcentratie hangt voornamelijk af van het celtype en de leeftijd van de celcultuur. Ter oriëntatie vindt u een volledig protocol in de technische informatiebrochure.
Na celdissociatie moet trypsine worden geïnhibeerd. Dit kan door middel van FCS of trypsine-inhibitoren.
