Zymolyase®, geproduceerd door een vloeistofkweek van Oerskovia xanthineolytica, heeft een sterke lytische activiteit tegen levende celwanden van verschillende stammen gistcellen. Daarom wordt het veelvuldig gebruikt voor de productie van protoplasten of sferoplasten. Het essentiële enzym voor de lytische activiteit van Zymolyase® is β-1,3-glucaan-laminaripentaohydrolase. Het hydrolyseert lineaire glucosepolymeren met β-1,3-bindingen en brengt specifiek laminaripentaose vrij als belangrijkste en kleinste producteenheid.
Andere bevestigde nevenactiviteiten: β-1,3-glucanase, protease, mannanase; incidenteel sporen van andere hydrolytische activiteiten (bijv. amylase, fosfatase).Stabiliteit: Bij 30 °C gaat na 3 maanden 70 % van de lytische activiteit verloren. Optimumtemperatuur en pH: bij pH 7,5 - 35 °C voor de lyse van levensvatbare gistcellen, bij pH 6,5 - 45 °C voor de hydrolyse van gistglucaan.
Eén eenheid lytische activiteit is gedefinieerd als de hoeveelheid enzym die een daling van 30 % in extinctie bij 800 nm veroorzaakt bij gebruik van 6 mg bakkersgist als substraat in fosfaatbuffer (pH 7,5) bij 25 °C. De lytische activiteit varieert afhankelijk van de specifieke giststam, het groeistadium van de gist en de kweekomstandigheden.
Zymolyase® 20T wordt bereid door precipitatie met ammoniumsulfaat.
