Substrat populaire des peroxydases. La DAB est utilisée pour la détection biochimique et histologique des peroxydases endogènes comme la catalase et la cytochrome oxydase. De plus, c'est le substrat le plus courant pour la peroxydase de raifort (HRP) dans les tests histochimiques et cytologiques. La DAB est oxydée par la HRP, formant un précipité brun, insoluble dans les solvants aqueux et organiques. Ce précipité peut être détecté en lumière visible et ne se décolore pas lors d'un stockage à long terme.
Concentration de travail : 0,05-0,1 % (0,5-1 mg/ml) de DAB dans un tampon (PBS, TBS, pH 7,0-7,6) contenant 0,01 % de peroxyde d'hydrogène. Toujours utiliser une solution de travail fraîchement préparée. Pour l'oxydation, des concentrations élevées de peroxyde d'hydrogène frais sont nécessaires.
