Les kits de modification PCR ont été développés pour la production de séquences d'ADN marquées par fluorescence. Lors de l'élongation de brin en PCR, la polymérase éthynyl contenue dans le kit incorpore des nucléotides modifiés par alcyne (EdUTP) dans le nouveau brin en plus des dNTP natifs. Il en résulte des produits PCR modifiés, qui peuvent ensuite être facilement marqués avec des colorants fluorescents à l'aide de la réaction Click. Les séquences marquées peuvent ensuite être utilisées pour la détection lors d'expériences d'hybridation ultérieures.
