Réduit la croissance des bactéries Gram-positives en inhibant l'initiation de la synthèse de l'ADN par liaison aux sites de liaison de l'ATP de la gyrase et de la topoisomérase. La novobiocine interagit également avec le site de liaison de l'ATP C-terminal de HSP90, inhibant ainsi sa fonction de chaperon.
Convient également comme additif aux milieux nutritifs microbiologiques. L'ajout de 40 mg/l de novobiocine lors de la détection sélective des bactéries coliformes sur le bouillon Brilliant Green Bile (Art. N° CL06.1) réduit efficacement la croissance de la flore accompagnatrice (Alcaide et al., 1982, J. Appl. Bact. 53:143-6).
