Application

Immunocytochimie (ICC) : dépend de l'essai. Immunohistochimie (IHC) - congelé : 1:100-1:200 (250-500 ng/ml). Immunohistochimie (IHC) - paraffine : 1:100-1:200 (250-500 ng/ml ; traitement par micro-ondes recommandé). Western Blot (WB) : dépend de l'essai.

Analyse IHC de cerveau de souris à l'aide d'un anticorps anti-GFAP. L'IHC a été réalisée sur des coupes de tissu fixées au formol et incluses dans la paraffine. Les échantillons ont été déparaffinés avec du xylol et de l'éthanol, suivis d'une récupération d'antigène induite par la chaleur avec un tampon citrate 10 mM. Après préparation, le tissu a été bloqué avec du sérum normal pendant 20 min à température ambiante. L'anticorps primaire anti-Protéine Acide Gliale Fibrillaire souris monoclonal, GF 12.24 (Réf. 690011) a été dilué dans du PBS (concentration d'anticorps 250 ng/ml) et incubé à 4°C pendant la nuit. L'anticorps secondaire ImmPRESS HRP anti-IgG de souris a été incubé pendant 20 min à température ambiante. Les lames ont été incubées avec une solution de DAB jusqu'à ce qu'une coloration brune soit visible et avec de l'hématoxyline pendant quelques minutes. L'image 20x a été acquise à l'aide d'un microscope (avec l'aimable autorisation de J. Hess, Hôpital Universitaire de Heidelberg).
