Le RPMI-1640 a été développé pour la culture de leucocytes humains normaux et néoplasiques et est basé sur des quantités modifiées d'acides aminés et de vitamines. Le milieu RPMI-1640 est très bien adapté à la culture d'un large éventail de lignées cellulaires de mammifères en suspension et de lymphocytes primaires, de tous les leucocytes, des lymphocytes B et T, ainsi que des cellules de myélome et d'hybridome.
En règle générale, du SVF est ajouté aux milieux RPMI. Cependant, il a été démontré que les milieux RPMI permettent également la croissance de plusieurs lignées cellulaires en l'absence de sérum.
Cette modification du RPMI contient de la L-glutamine, mais pas de pyruvate.
