Snel en veilig proces voor de analyse van coliforme bacteriën in het algemeen, en specifiek E. coli in water.
De test is ook geschikt voor de analyse van troebele watermonsters en watermonsters die zwevende stoffen bevatten. Bacteriën die door gechloreerd water gestrest zijn, kunnen over het algemeen ook worden aangetoond. Het EC Blue-systeem bespaart tijd en voorbereidend werk omdat het reagensmengsel in steriele testflessen voor direct gebruik wordt geleverd. De aanwezigheid van E. coli kan eenvoudig worden gedetecteerd door het optreden van fluorescentie onder UV-licht (zie EC-Blue Mini UV-lamp art. nr. KK49.1). Ongeveer 95% van alle E. coli-stammen produceert het enzym β-glucuronidase, dat reageert met een enzymsubstraat in EC Blue om een fluorescerende kleurstof vrij te maken. Kwantificering is mogelijk met behulp van EC Blue Quant/MPN (art. nr. 0199.1).
Giet 100 ml van het testwater in een steriele container en los de inhoud van een portiezakje EC Blue 100P-poeder erin op. Het ontstaan van een blauwe tot blauwgroene kleur na incubatie bij 35 °C duidt op de aanwezigheid van coliforme bacteriën in het water. Deze kleurverandering is te wijten aan een enzymatische kleurreactie van de coliform-specifieke galactosidase met het chromogene substraat X-Gal.
