Deze HPLC-kolom is gepakt met een polaire aminopropyl-modificatie, gebaseerd op NUCLEODUR®-silicagel van hoge zuiverheid. Deze HPLC-fase is in staat om een duidelijk herkenbaar ander retentiegedrag te genereren in vergelijking met puur alkyl-gefunctionaliseerde oppervlaktemodificaties. Naast cyano-modificaties behoren amino-modificaties tot de meest gebruikte polaire silicafasen. Aminofasen kunnen zowel in de RP-modus worden gebruikt met waterig-organische eluentmengsels als in de NP-modus, bijv. met hexaan als mobiele fase. Ook anionenuitwisselingschromatografie van anionen en organische zuren met behulp van gebruikelijke buffers (bijv. acetaat of fosfaat) in combinatie met organische modificatoren (bijv. acetonitril) is mogelijk. Scheidingsmechanisme gebaseerd op polaire interacties, hydrofobe interacties en zwakke ionenuitwisselingsinteracties.
