Weefsels die vast materiaal bevatten, zoals botten of keratine, moeten worden ontkalkt voor inbedding in paraffine. Gewoonlijk worden de monsters behandeld met zuren van verschillende concentraties en mengsels.
Een bijzonder milde methode is ontkalking met EDTA-oplossing.
De oplossing bevat trichloorazijnzuur en formaline en heeft daarom ook fixerende eigenschappen. Niettemin raden wij aan de monsters voldoende te fixeren alvorens ze te ontkalken, omdat trichloorazijnzuur morfologische veranderingen in niet-gefixeerd weefsel kan veroorzaken.
De duur van het ontkalkingsproces hangt af van de grootte en het type monster. Kleine botten (diameter enkele mm) hebben 1-2 dagen nodig voor volledige ontkalking. De ontkalker standaard is niet geschikt voor immunologische toepassingen. Wij adviseren onze ontkalker mild (art.nr. 6484) op basis van EDTA.
