Weefsels die vaste bestanddelen bevatten, zoals botten of keratine, moeten vóór het inbedden in paraffine worden ontkalkt. Meestal worden de preparaten behandeld met zuren in verschillende concentraties en mengsels.
Een bijzonder milde methode is ontkalking met EDTA-oplossing.

De oplossing bevat 25 % EDTA.
Het ontkalkingsproces neemt meer tijd in beslag dan zuurontkalking (zie Decalcifier standard, art.nr. 6483), bijv. botbiopten hebben ca. 3-4 dagen nodig. De procedure kan worden versneld met behulp van een magneetroerder. De ontkalking vindt plaats in een zwak basisch milieu. Nucleïnezuren en enzymen worden niet aangetast; daarom is de oplossing ook geschikt voor immunologische toepassingen.
