RPMI-1640 werd ontwikkeld voor het kweken van normale en neoplastische humane leukocyten en is gebaseerd op aangepaste hoeveelheden aminozuren en vitaminen. RPMI-1640 medium is uitermate geschikt voor het kweken van een breed scala aan zoogdier-suspensiecellijnen en primaire lymfocyten, alle leukocyten, B- en T-lymfocyten, evenals myeloom- en hybridoomcellen.
Over het algemeen wordt FBS toegevoegd aan RPMI-media. Het is echter aangetoond dat RPMI-media ook verschillende cellijnen ondersteunen in afwezigheid van serum.
Deze modificatie van RPMI bevat L-glutamine, maar geen pyruvaat.
