ROTI®Load 1 (reducerend) evenals ROTI®Load 2 en 3 (niet-reducerend) zijn speciale gellaadbuffers voor proteïne-gelelektrofores. Ze zijn ontworpen om uw eiwitten te beschermen tijdens de monstervoorbereiding en om gevoelige peptidebindingen te stabiliseren. Eiwitafbraak wordt door deze buffersystemen efficiënt voorkomen. ROTI®Load zijn 4x geconcentreerde, gebruiksklare oplossingen die direct voor het laden van monsters kunnen worden gebruikt.
Gemodificeerde, niet-reducerende proteïne-monstergellaadbuffer voor gels onder denaturerende of natieve condities.

ROTI®Load proteïne-gellaadbuffers worden gewoonlijk gebruikt als 1x geconcentreerde oplossing. ROTI®Load kan aan het voorbereide monster worden toegevoegd of de geprecipiteerde eiwitten kunnen direct in ROTI®Load worden opgelost. Optioneel kan het met ROTI®Load gemengde monster gedurende 5 minuten tot 95 °C worden verhit om waterstofbruggen te verbreken en de peptideketens uit te rekken.
Tijdens de monstervoorbereiding en verhitting voorkomt ROTI®Load 3 (LDS) efficiënt de afbraak van uw eiwitten en stabiliseert het tevens de pH-waarde tijdens de loop. Deze gellaadbuffer is gebaseerd op een met triethanolamine gebufferde, gebruiksklare oplossing, waarin het standaard natriumdodecylsulfaat (SDS) is vervangen door lithiumdodecylsulfaat (LDS). Bij lage temperaturen is LDS veel beter oplosbaar dan SDS. Hierdoor treedt er geen kristallisatie van LDS op en worden de eiwitstabiliteit en de bandresolutie aanzienlijk verbeterd. Bovendien is de formulering van ROTI®Load 3 (LDS) geoptimaliseerd voor kant-en-klare gels, in het bijzonder Q-PAGE™-gels, maar ook voor kant-en-klare gels van andere standaardleveranciers. Voor reducerende condities kan ROTI®Load 3 (LDS) worden aangevuld met 20 µl/ml DTT-oplossing (1 M) of met β-mercapto-ethanol.
