Questions about Chromatography equipment

What is chromatography?

Chromatography separates a mixture by letting it travel between a mobile phase and a stationary phase. A compound that prefers the stationary phase lags behind. A compound that prefers the mobile phase moves ahead. The result is a sequence of peaks, or of spots, instead of one mixed sample. The name and the definition are set by IUPAC. Identity of the molecule is looked up on PubChem, and spectra and physical data on the NIST Chemistry WebBook.

Four techniques cover almost every order. In HPLC the mobile phase is a liquid pushed through a packed column: see HPLC columns and accessories. In GC it is a gas passing a coated capillary: see GC columns and accessories. In TLC the mobile phase climbs a coated plate by capillary action: see TLC. SPE uses the same partition to clean a sample before the chromatogram is drawn, not to draw it: see SPE and sample preparation. The pump, the detector and the oven are instruments. This catalogue is the column, the plate, the cartridge and the vial.

How do I specify a column, a cartridge or a vial?

An HPLC column is decided by the stationary phase, the particle size, the pore, the length and the internal diameter. A GC column is decided by the stationary phase, the film thickness, the length and the internal diameter. An SPE cartridge is decided by the sorbent, the bed mass and the tube volume. A vial is decided by the volume, the neck and the septum, on vials, caps and septa. The liquid of the mobile phase is chosen from solvents. None of these numbers is a guess: they come from the method the bench already runs.

Check the substance on PubChem and the naming on IUPAC. Physical data sit on the NIST Chemistry WebBook. When the method cites a pharmacopoeia, use the European Pharmacopoeia or USP. Reading on method validation is published by Eurachem. The safety data for a solvent is the manufacturer sheet on the product page, not a rewritten hazard block.
