HEPES is a versatile buffer, which is zwitterionic over a broad pH-range, interesting for biological assays, and, therefore, doesn’t pass through biological membranes. In contrast to NaHCO3 buffers, it also doesn’t release any CO2 which affects the surroundings and has to be compensated. Hence, HEPES is particularly adapted for cell- and tissue cultures. HEPES reacts with DEPC. It's recommended to use DNase/RNase/Protease free HEPES BioScience-Grade as soon as molecular biological applications like PCR, extraction, isolation, hybridization etc. are to be performed.
