DAB-substraatkits voor detectie van peroxidase (HRP)-activiteit op immunoblots, immuno-histo- en cytochemische preparaten en tijdens in situ hybridisatie.
ROTI®DAB Kit bevat substraat, bufferoplossing en activeringsreagens, dus inclusief alle drie de oplossingen die nodig zijn voor de HRP-detectiereactie. De toepassing is zeer eenvoudig - alle drie de kitoplossingen worden gelijkmatig verdund in water, waarna het blotfiltermembraan of de preparaten worden geïncubeerd in deze substraatoplossing. Aangezien de oplossingen worden geleverd in druppeldispensers, is het doseren zeer handig.
DAB wordt geoxideerd door HRP, waarbij een bruine precipitaat ontstaat die onoplosbaar is in waterige en organische oplosmiddelen. Deze precipitaat kan worden gedetecteerd in zichtbaar licht en verbleekt niet bij langdurige bewaring.
Toevoeging van de DAB Metal Enhancer (Art. Nr. 9204) verdubbelt de signaalsterkte. Preparaten kunnen permanent worden ingesloten in hydrofiel of, na dehydratie, in hydrofoob insluitmiddel.
DAB-oplossing (Art. Nr. 9869), DAB-buffer (Art. Nr. 9870) en Activeringsreagens (Art. Nr. 9871) in druppelflessen. De inhoud van deze kit kan niet apart worden gekocht.
