Agent fortement chaotropique qui détruit puissamment la structure tridimensionnelle des protéines, entraînant leur dénaturation. Le thiocyanate de guanidinium (iso) contient de puissants groupes anioniques ainsi que cationiques, qui forment de fortes liaisons hydrogène. Il peut donc être utilisé pour rompre les liaisons disulfure des protéines ainsi que pour perturber les interactions hydrophobes, et est ajouté lors de l'extraction de l'ARN afin d'inhiber l'activité des ARNases.
